Resumo: Esta publicação tem por objetivo descrever a história, a composição da Coleção de trabalho de Microrganismos Patogênicos a Caprinos e Ovinos, e de que forma ela tem conservado, caracterizado, documentado e utilizado seus isolados. A coleção foi criada em 2012 e atualmente é composta por 141 cepas bacterianas e seis cepas virais, pertencentes às espécies Corynebacterium pseudotuberculosis, Staphylococcus aureus e ao gênero Lentivirus. Esses isolados são utilizados para produção de antígenos para imunodiagnósticos, em diagnósticos moleculares e na avaliação de resistência a antimicrobianos e ação de anti-sépticos e antimicrobianos. Visando o melhoramento genetico da sacaca, foi instalado um Banco Ativo de Germoplasma (BAG) na Embrapa Amazonia Ocidental. Estao sendo feitas coletas e multiplicacao vegetativa de acesso de sacaca de diversos locais da Amazonia. O BAG-Sacaca visa nao somente a conservacao do germoplasma, mas o estudo da variabilidade genetica entre acesso, para selecao de genotipos superiores para producao de biomassa (folhas), e com maior potencial de producao de oleos essenciais. Developing an in vitro optimized protocol to sweet potato landraces conservation Agronomy

This study aimed to develop a protocol for the in vitro conservation of 30 accessions of sweet potato collected in the northern region of Rio de Janeiro State, Brazil, aiming to extend the period between subcultures for this species. An in vitro minimal growth experiment was established with accession UENF 1931 in a factorial arrangement consisting of four concentrations of mineral salts in MS medium and four concentrations of sucrose, at two temperatures. Development and characterization of polymorphic microsatellite markers for the sweet orange (Citrus sinensis L. Osbeck) Genet. Mol. Biol.

The aims of this study were to develop simple sequence repeat markers (SSRs or microsatellite markers) in citrus and to evaluate the efficiency of these markers for characterization of sweet orange. We developed SSRs from a genomic library of 'Pêra IAC' sweet orange enriched for AG/TC, GT/CA, TCA/AGT and AAC/TTG sequence repeats. We selected 279 sequences from which 171 primer pairs were designed of which 113 with the best banding patterns were selected. Characterization of sweet orange microsatellite loci revealed that AG/TC was the most abundant (69%) microsatellite class isolated, followed by GT/CA (15.9%), TCA/AGT (8%) and AAC/TTG (6.2%). Jacaranda decurrens (Bignoniaceae) is an endemic species of the Cerrado with validated antitumoral activity. The genetic diversity of six populations of J. decurrens located in the State of São Paulo was determined in this study by using molecular markers for randomly amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP). Following optimization of the amplification reaction, 10 selected primers generated 78 reproducible RAPD fragments that were mostly (69.2%) polymorphic. Germplasm characterization of three jabuticaba tree species Rev. Bras. Frutic.

The purpose of this study was to characterize cultivated genotypes of three jabuticaba species (Plinia cauliflora, P. trunciflora, and P. jaboticaba). Phenology and fruit growth, as well as leaf, flower and fruit traits were evaluated. Variability in all traits was observed among genotypes of the three jabuticaba species. The trait peduncle size is indicated for differentiation of the three species under study. The leaf and fruit sizes of the genotypes P. trunciflora 3, P. trunciflora 4, P. trunciflora 5 and P. jaboticaba 1 differ from those described in the literature for these species, indicating the formation of ecotypes. Jabuticaba fruit skin contains high anthocyanin and flavonoid concentrations, with potential use in food and pharmaceutical industries.

ABSTRACT: Adoption of resistant cultivars is the primary measure used to control anthracnose stalk rot. The goal of this study was to identify maize-resistant genotypes to anthracnose stalk rot, which are similar to the hybrid 2B710. Experiments were performed at Embrapa Maize and Sorghum experimental fields in Brazil. The first experimental trial evaluated 234 maize lines as well as two commercials hybrids, BRS1010 (susceptible) and 2B710 (resistant). In vitro conservation of Piper aduncum and Piper hispidinervum under slow-growth conditions PAB

The objective of this work was to evaluate in vitro storage of Piper aduncum and P. hispidinervum under slow-growth conditions. Shoots were stored at low temperatures (10, 20 and 25°C), and the culture medium was supplemented with osmotic agents (sucrose and mannitol - at 1, 2 and 3%) and abiscisic acid - ABA (0, 0.5, 1.0, 2.0 and 3.0 mg L-1). After six-months of storage, shoots were evaluated for survival and regrowth. Low temperature at 20ºC was effective for the in vitro conservation of P. aduncum and P. hispidinervum shoots. In vitro cultures maintained at 20ºC on MS medium showed 100% survival with slow-growth shoots. Cassava (Manihot esculenta Crantz) belongs to the Euphorbiaceae family, and is widely cultivated in Brazil. The objective of this study was to evaluate the isoenzymatic variability of 200 cassava accessions from the germplasm bank of Embrapa Amazonia Oriental. Seven groups were formed according to their origin: 1-Amazonas, 2-Amapa, 3-Bahia, 4-Para, 5-Rondonia, 6-Various, for accessions with a maximum of three individuals per place of origin, and 7 - Accessions of indefinite origin.
